A Study on the Expression of Alpha2,6-sialyltransferase in CIN? ...

[Abstract]

Objective:Expression of alpha2,6-sialyltransferase(ST6GalI) was checked in CINIII, invasivesquamous cell carcinoma of cervix uteri(SCC), SCC before and afeter neoadjuvantchemotherapy(NACT), to assess the association of the expression of ST6GalI andchemosensitivity.Methods:1?Expression of ST6GalI in normal cervix samples(n=10), CINIII samples(n=20)and SCCsamples including FIGO stage IB2(n=10), IIA(n=14) and IIB(n=12) wereexamined by Elivision two-steps immunohistochemistry method, analysidng theexpression of ST6GalI correlated with the clinical factors including age, clinical stage,grade of differentiation and lymph node metastasis. All tumor sizes of cervical cancerwere equal or greater than4cm. The positive effect of ST6GalI expressed in cellmembrance and cytoplasm, browm-yellow or brown-chocolate granular substance.2?Expression of ST6GalI in the patients of SCC before-after NACT who wereanterior-posterior self-controlled pairing(n=36) were investigated by Elivision two-stepsimmunohistochemistry method, analysing the effect of chemotherapy related with theexpression of ST6GalI for before-after neoadjuvant chemotherapy as well as the clinicalfactors including age, clinical stage and grade of differentiation.3?Expression of ST6GalI mRNA levels in the patients of invasive cervicalsquamouscell carcinomas before-after NACT who were anterior-posterior self-controlled pairing(n=10) were examined by real-time fluorescent quantitative polymerase chain reaction(FQ-PCR).4?SPSS13.0was used for statistical analysis software to analyse the experimentaldata, and adopted consider a=0.05as the detection standard. Results:1?The positive expression ratio of ST6GalI in normal cervix, CINIII and SCC was0%,30%(6/20) and61.9%(23/36), respectively, and the positive expression of ST6GalIwith the degree of cervical disease was positive correlation. The expression of ST6GalIin SCC was significantly higher than in normal cervix and CINIII(P<0.05, respectively).2?The expression of ST6GalI in SCC was not significantly correlated with age,clinical stage, grade of differentiation and lymph node metastasis(P>0.05, respectively).3?Nine patients achieved a complete remission, sixteen patients achieved a partialremission, ten patients achieved a stable disease or no change, and one patient achieved aprogressive disease. Total clinical effective ratio was69.4%(25/36).4?The effect of chemotherapy was not correlated with age, clinical stage and gradeof differentiation(P>0.05, respectively). The positive expression ratio of ST6GalI withNACT(before and after) was61.9%(23/36)and16.7%(6/36), respectively, which wassignificantly descended after NACT (P<0.05). In clinical curative effect group, positiveexpression of ST6GalI before vs. after NACT was8%(2/25)vs.(72%,18/25),(P<0.05).In clinical ineffective group, positive expression of ST6GalI before vs, after NACT was36.4%(4/11), vs.(45.5%,5/11),(P>0.05).5?The relative expression of ST6GalI mRNA before and after NACT was0.42?0.61(X?S)and0.04?0.06((X?S), respectively. ST6GalI mRNA expression wassignificantly decreased in SCC with after NACT vs.before NACT by relative quanti-tative2???Ctmethod(P<0.05, Wilicoxon signed-rank sum test of paired samples).Conclusion:1?The positive expression of ST6GalI with the degree of cervical disease waspositive correlation, and its expression is not significantly correlated with age, clinicalstage, grade of differentiation and lymph node metastasis;2?The expression of ST6GalI with the effective chemotherapy is significantlydecreased, and its expression with the ineffective chemotherapy has not notable change.
